Regulatory expression of Co-inhibitory receptors by Type 1 Interferon transcriptional network in human T cells

bioxone November 2, 2020November 2, 2020

Rohit Bhattacharjee, Amity University Kolkata

Although inhibition of T cell co-inhibitory receptors has brought about cancer therapy, mechanisms governing their expression on human T cells have not been explained. Type 1 interferon (IFN-I) regulates T cell immunity in viral infection, autoimmunity, cancer, and may enhance induction of T cell exhaustion in chronic viral infection. To enable the construction of dynamic transcriptional regulatory networks high-temporal resolution mRNA profiling of IFN-I responses were done to uncover three temporal transcriptional waves.

After disturbance of the key transcription factors on human primary T cells, it revealed both canonical and non-canonical IFN-I transcriptional regulators and recognised unique regulators controlling the expression of co-inhibitory receptors. To provide direct in vivo evidence for the role of IFN-I on co-inhibitory receptors, single-cell RNA-sequencing in subjects infected with SARS-CoV-2 was performed, where the viral load was strongly associated with T cell Type 1 Interferon (IFN-I) signatures. It was found out that the dynamic IFN-I response in vitro closely mimicked T cell characteristics with acute IFN-I linked viral infection, having high LAG3 and low TIGIT expression.

Lastly, the gene regulatory network identified SP140 as a key regulator for differential LAG3 and TIGIT expression. The construction of IFN-I induced co-inhibitory regulatory networks with identification of unique transcriptional factors controlling their expression may provide targets for enhancement of cancer immunotherapy, infectious diseases, and autoimmunity. Conclusively, this approach identifies the cytokine signals and regulatory mechanisms driving the expression of humans’ co-inhibitory receptors, providing a pathway to comprehensively capture the dynamics of their human expressions.

Immortality- a fate worse than death?

bioxone November 2, 2020

Ayooshi Mitra, Amity University Kolkata Immortality is defined as eternal life i.e. being untouched by death, also acknowledged as eternal existence. Why do living beings gradually decay when they grow older? Senescence is the steady degeneration of cells or more appropriately of the entire organism ultimately. Organismal senescence leads to an increase in mortality rate […]

iBlastoids: Reprogrammed skin cells as a Human Embryo

bioxone March 23, 2021March 23, 2021

Devyani Goswami, Amity University Kolkata José M. Polo, PhD, a professor of anatomy and developmental biology at Monash University, discovered a way to generate induced blastoids or iBlastoids that imitates the natural human blastoids; made up of reprogrammed skin cells. Molecular and functional assays were performed to demonstrate that the structures were blastocyst-like structures that […]

AISHILA KAR, AMITY UNIVERSITY KOLKATA Behavioural states of arousal are maintained by a system of nuclei spread all over the brain.  Endogenous sleep and general anaesthesia are well-defined states that share similar traits.  Especially to neuroscience is the loss of consciousness that goes through both states. Hypothalamic preoptic area (POA) neurons are intimately connected in […]

Coronavirus gene remains in human DNA after the infection is over

bioxone May 20, 2021May 20, 2021

Rohit Bhattacharjee, Amity University, Kolkata A group of prominent researchers has multiplied down on its controversial speculation that hereditary pieces of the pandemic Coronavirus can integrate into our chromosomes and stick around long after the infection is over. If they are correct, the additions could clarify the uncommon finding that individuals can recuperate from COVID-19 […]
