Breast cancer is among the second largest cause of deaths among women related to cancer. The development of breast cancer is a multi-step process that involves numerous cell types and its treatment remains shrouded with many challenges. Triple-negative breast cancers (TNBC) is accounted for 10-15% of all breast cancers. Triple-negative breast cancer differs from other breast cancers and is referred as to an aggressive type wherein the affected cancer cells are devoid of estrogen, progesterone receptors, and HER2 protein. It is cancer that tests negative for all of the above-mentioned proteins. It is very difficult to diagnose TNBC with a mammogram due to its rapid growth. Treatment is limited but through targeted therapeutic regimes the horizon was been broadened. About 80% of the patients suffering from TNBC do not achieve any response from chemotherapy, however genetic counselling targeted treatment for individuals has been beneficial. Pembrolizumab monotherapy has recently shown great response in antitumor activities and other important therapies include Talazoparib, Veliparib, Pembrolizumab with eribulin, and Atezolizumab plus nab-paclitaxel. Stakeholders and researchers duly ratified that unmet needs remain in the treatment of TNBC.
Specifically, a high need exists for more efficient and efficacious therapies, particularly in the management of patients who deteriorate early or are resistant to chemotherapy. Furthermore, there is an urge to determine why immune reactions are increasingly suppressed as patients get prone to their disease course with later lines of therapeutics, which will be imperative for future considerations.
