Behavioural states of arousal are maintained by a system of nuclei spread all over the brain. Endogenous sleep and general anaesthesia are well-defined states that share similar traits. Especially to neuroscience is the loss of consciousness that goes through both states. Hypothalamic preoptic area (POA) neurons are intimately connected in modulating automatic and anaesthetic-induced transmute in arousal. However, recent work proposes that POA GABAergic or glutamatergic neurons able to regulating endogenous sleep fail to the disappearing of anaesthesia.
The potent results of Tac1 activation thus work to hold up the waking state over endogenous as well as anaesthetic-induced damage of arousal. Chemogenetic inhibition, although incomplete, but did not influence arousal state, suggesting the POA Tac1 neurons are enough, but perhaps not essential, for wakefulness. On other hand, to a previously published report, it shows that chemogenetic activation of POA Tac1 neurons destroys both the non-rapid eye movement (NREM) and rapid eye movement (REM) sleep, strongly consolidating the waking state for hours, even at the time of elevated sleep drive.
Furthermore, chemogenetic activation of Tac1 POA neurons maintains the waking state against both isoflurane- and sevoflurane-induced unconsciousness. Tac1-activated mice display a partial resistance to entering isoflurane anesthesia while possessing more ability to leave both isoflurane- and sevoflurane-induced unconscious states.
We conclude that POA Tac1 neurons can potently strengthen arousal both against endogenous and drug-induced unconscious states. These results that the shared anaesthetic hypnosis, which posits that anaesthetics exert their hypnotic effects in part by acting on endogenous sleep-wake neuronal circuitry and that independent modulation of the same neurons in these circuits influences the anaesthetic state. Although, recent work showing that GABAergic or glutamatergic POA neurons that modulate sleep and wake do not influence isoflurane induction. Further recognition of the hypothalamic microcircuitry controlling sleep and wakefulness with the capacity to impact drug-induced states of unconsciousness is required.
