Biomarker TIICs benefitting the prognosis in osteosarcoma patients

Osteosarcoma is a rare malignant bone tumour found in adults and children. It is characterized by early lung metastasis and poor prognosis that seriously the physical and mental health of patients. Chemotherapy and surgery include the main clinical treatment however, recurrence of metastasis remains the poor prognosis. Advances in the study of the tumour microenvironment (TME) opened new directions for the study of osteosarcoma. Recent studies showed that Tumour Infiltrating Immune Cells (TIICs) are associated with osteosarcoma patients’ clinical outcomes. 

CIBERSORTX algorithm was used for calculating the proportion of 22 TIIC types in osteosarcoma samples. Kaplan-Meier curves were drawn to find out the prognostic value of 22 TIIC types. Results revealed that macrophages account for a large proportion of 22 immune cell types, Kaplan-Meier curves indicated naïve B cells and monocytes are associated with poor prognosis of osteosarcoma. B lymphocytes exert a two-way role in tumour – positively regulating tumour immune process by secreting cytokines, antibody production, and negatively regulating tumour immune process by inhibition of proliferated immune-activated T cells.

Also, a nomogram model was created based on immune risk score model which quantified the probability of an event individually and accurately by integrating multiple predictors that are based on multi-factor analysis which showed that TIICs are very closely associated with osteosarcoma prognosis and the immune risk score model created based on TIICs is reliable in osteosarcoma prognosis prediction.
