Salivary duct carcinoma (SDC) is one of the rarest incursive subtypes of salivary gland cancer that microscopically resembles high-grade ductal carcinoma of the breast, with both in situ and invasive pattern. Ordinary chemotherapy and radiation have just restricted viability for the metastatic SDC2, thus treatment is needed against this unique tumor type.

Multi-omic profiling of the SDC has been conducted to describe the genomic alternations prevalent in the disease. The findings from whole genome sequencing and transcriptome sequencing of the original discovered cohort of 10 SDC samples were supported by 32 SDC samples.

85 somatic exonic mutations were found per sample. A mutational signature of BRCA and APOBEC/AID was found in the cancer cohort. There are several genes such as TP53, RBI, MAD4, HRAS, APC, PIK3CA, and GNAQ which were somatically altered in SDC. MYB-NHSL1 – a novel fusion gene, generated by rearrangement of the genome was also noted.
